allelic dimorphism of the plasmodium falciparum erythrocyte binding antigen-175 (eba-175) gene in the south-east of iran

abstract
background: the erythrocyte binding antigen 175 kda (eba-175) gene is located on chromosome 7. it encodes protein that binds to specific receptor glycophorin a on the erythrocyte surface during invasion. it has a dimorphic nature (fcr3 and camp). this study was designed to determine the distribution of eba-175 alleles of plasmodium falciparum in the southeast of iran methods: we used the nested pcr method with specific primers, which improves the two fragments of the eba-175 gene. sixty eight microscopically positive blood samples were collected from the infected falciparum malaria subjects in the southeast of iran. results: in this study which marks the first one in iran, camp strains (714 bp) and fcr-3 strains (795 bp) were found in 14 (37.8%) and 23 (62.2%) in the originally iranian subjects and in 10 (32.3%) and 19 (61.3%) pakistani infected migrants respectively. two migrant cases (6.4%) had mix camp/fcr-3 infection. conclusion: the two fragments of dimorphic eba-175 gene were observed and the fcr-3 allele was more prevalent in iran. there was no significant correlation between one of the eba-175 alleles and the subject group in the mentioned region. this distributional pattern should be considered in designing to control p. falciparum malaria in the region.

Genotyping of C and F Regions of Plasmodium Falciparum EBA-175 in South-East of Iran
Abstract Background and Objective: The Plasmodium falciparum EBA-175, via Sialic acid dependent glycophorin A, binds to red blood cells and thus plays a critical role in cell invasion. Some part of second allele in its gene encoding in FCR-3 (Section F) and CAMP (Section C) can be found. This study aimed to determine the prevalence of Plasmodium falciparum EBA-175KD alleles in southeastern I...
full textDistribution of erythrocyte binding antigen 175 (EBA-175) gene dimorphic alleles in Plasmodium falciparum field isolates from Sudan
BACKGROUND The Erythrocyte Binding Antigen (EBA) 175 has been considered as one of the most important Plasmodium falciparum (P. falciparum) merozoite ligands that mediate invasion of the erythrocytes through their sialated receptor: Glycophorin A (GPA). The effect of the EBA 175 dimorphic alleles (F and C) on the severity of the disease is not yet fully understood. Therefore this study was desi...
full textCharacterization of a Plasmodium falciparum erythrocyte-binding protein paralogous to EBA-175.
A member of a Plasmodium receptor family for erythrocyte invasion was identified on chromosome 13 from the Plasmodium falciparum genome sequence of the Sanger Centre (Cambridge, U.K.). The protein (named BAEBL) has homology to EBA-175, a P. falciparum receptor that binds specifically to sialic acid and the peptide backbone of glycophorin A on erythrocytes. Both EBA-175 and BAEBL localize to the...
full textEvidence for two-stage binding by the 175-kD erythrocyte binding antigen of Plasmodium falciparum
Plasmodium falciparum malaria merozoites invade human erythrocytes bearing sialic acid in a multistage process involving the sialic acid-dependent binding of a malaria molecule, the 175-kD erythrocyte binding antigen (EBA-175). We show here that after the initial interaction of EBA-175 with its sialic acid-containing erythrocyte determinant, endogenous proteases can cleave EBA-175 to 65-kD frag...
full textStructural Basis for the EBA-175 Erythrocyte Invasion Pathway of the Malaria Parasite Plasmodium falciparum
Erythrocyte binding antigen 175 (EBA-175) is a P. falciparum protein that binds the major glycoprotein found on human erythrocytes, glycophorin A, during invasion. Here we present the crystal structure of the erythrocyte binding domain of EBA-175, RII, which has been established as a vaccine candidate. Binding sites for the heavily sialylated receptor glycophorin A are proposed based on a compl...
full textAnalysis of human antibodies to erythrocyte binding antigen 175 peptide 4 of Plasmodium falciparum.
BACKGROUND The IgG1 and IgG3 antibodies are considered cytophilic and protective against Plasmodium falciparum, whereas IgG2 and IgG4 are thought to block protective mechanisms. OBJECTIVES The main objective was to measure antibodies directed against erythrocyte binding antigen-175 (EBA-175) peptide 4 and analyze the relationship between such antibodies and clinical malaria attack. METHODS ...
